PENGEMBANGAN METODE KLT VIDEO DENSITOMETRI UNTUK ANALISIS MIKONAZOL NITRAT DAN HIDROKORTISON ASETAT DALAM SEDIAAN KRIM

Infeksi jamur pada kulit dapat diatasi dengan sediaan krim yang mengandung mikonazol nitrat (MN) dan hidrokortison asetat (HA). Penetapan kadar kedua zat tersebut dapat dilakukan secara simultan menggunakan beberapa metode seperti KCKT, spektrofotometri UV dan FTIR, akan tetapi penggunaan metode KLT video densitometri belum pernah dilaporkan di literatur. Tujuan dari penelitian ini adalah untuk mengembangkan KLT video densitometri untuk analisis MN dan HA yang efektif dalam formulasi krim. Tahapan penelitian dimulai dari pembuatan larutan baku, optimasi sistem KLT, validasi metode analisis dan penetapan kadar MN dan HA pada sediaan krim. Sistem KLT terbaik untuk pemisahan yaitu menggunakan fase diam silika gel GF254 dan fase gerak campuran n-heksan:kloroform:metanol (6:3:1). Bercak pada plat KLT divisualisasikan di bawah lampu UV254, kemudian direkam menggunakan kamera digital, dan dianalisis secara kuantitatif menggunakan software ImageJ untuk menghitung luas area. Hasil penelitian menunjukkan nilai faktor selektivitas (α) sebesar 1,52. Nilai koefisien korelasi (r) untuk keduanya diatas 0,99. Nilai batas deteksi dan batas kuantitasi berturut-turut adalah 30,63 dan 102,10 bpj untuk MN, serta 12,12 dan 40,39 bpj untuk HA. Perolehan kembali sebesar 100,84±1,25% untuk MN dan 100,27±0,89% untuk HA, sedangkan nilai presisi (%SBR) untuk MN dan HA sebesar 0,647% dan 0,547%. Kadar MN dan NA dalam sediaan krim berturut-turut adalah 100,41±0,27% dan 101,15±0,18%. Metode KLT video densitometri dapat digunakan sebagai metode analisis alternatif yang sederhana, cepat dan andal dalam penetapan kadar zat aktif MN dan HA dalam sediaan krim kombinasi.
