UJI AKTIVITAS ANTIDIABETES EKSTRAK DAUN KERSEN (Muntingia calabura L.) PADA MODEL HEWAN UJI ZEBRAFISH DIABETES

Daun kersen (Muntingia calabura L.) diketahui mengandung flavonoid, seperti kuersetin, yang berpotensi antihiperglikemik. Penelitian ini bertujuan mengoptimasi pemodelan diabetes melitus (DM) pada zebrafish dan mengevaluasi aktivitas hipoglikemik ekstrak etanol daun kersen. Ekstrak distandardisasi melalui penetapan kadar air dan identifikasi KLT menggunakan pembanding kuersetin. Dua pendekatan model DM dievaluasi, yakni perendaman dalam glukosa 4% serta kombinasi glukosa 2% dengan pakan tinggi lemak. Uji hipoglikemik membandingkan kontrol normal, kontrol negatif (glukosa 4%), kontrol positif (metformin 1,65 mg/dL), dan tiga dosis ekstrak (50, 100, 150 mg/L) pada hari ke-0, 3, 5, 7, dan 14. Ekstrak memenuhi mutu awal (kadar air 3,69%; bercak KLT dengan Rf 0,346 ko-migrasi dengan kuersetin). Perendaman dalam glukosa 4% menghasilkan zebrafish hiperglikemia dengan kadar gula darah >1,5x dari nilai baseline, responsif terhadap metformin, dan bermortalitas rendah, sedangkan induksi dengan kombinasi pakan dan glukosa gagal karena mortalitas ±30%/hari. Pada pengujian hipoglikemik, kadar glukosa kontrol negatif meningkat hingga 227 ± 5,37 mg/dL (H14), sementara metformin menurunkannya menjadi 29,5 ± 2,43 mg/dL (H14; −73,5%). Ekstrak menurunkan glukosa secara bermakna pada seluruh dosis; dosis 150 mg/L mencapai 24,5 ± 1,05 mg/dL (−78,0%) dan lebih rendah daripada metformin, sedangkan dosis 50–100 mg/L berada pada 40,17–38,83 mg/dL (−63,9% s.d. −65,1%). Analisis ANOVA satu arah/Welch menunjukkan perbedaan signifikan antar-kelompok dan antar-waktu (p<0,001), dan uji lanjut Games–Howell mengonfirmasi bahwa seluruh dosis ekstrak berbeda bermakna dari kontrol negatif, dengan dosis 150 mg/L lebih efektif dibanding 50–100 mg/L pada H14. Temuan ini menegaskan bahwa model zebrafish glukosa 4% optimal untuk skrining antidiabetes dan bahwa ekstrak daun kersen memiliki aktivitas hipoglikemik signifikan yang mendukung pengembangannya sebagai kandidat obat antidiabetes alami.
