BIOKONVERSI LIMBAH MANGGA MENJADI ASAM SITRAT MELALUI FERMENTASI TERKONTROL Aspergillus niger ATCC 16404

Biaya substrat karbon merupakan komponen terbesar dalam produksi asam sitrat industri, sehingga diperlukan sumber alternatif yang ekonomis. Penelitian ini bertujuan mengevaluasi potensi limbah daging buah mangga (Mangifera indica L.) sebagai substrat alternatif bagi Aspergillus niger ATCC 16404. Fermentasi dilakukan melalui metode fermentasi media cair terendam (submerged fermentation) selama 108 jam menggunakan vertical shaker 150 rpm dengan variasi konsentrasi limbah (A: 0%, B: 25%, dan C: 50%). Parameter yang diukur meliputi kinetika pertumbuhan sel, konsumsi gula pereduksi, dinamika pH, dan kadar asam sitrat yang divalidasi dengan uji statistik ANOVA satu arah dan DMRT (α = 0,05). Hasil penelitian menunjukkan bahwa peningkatan konsentrasi limbah mangga meningkatkan akumulasi produk secara sangat signifikan (F = 21,93; p < 0,001). Variasi konsentrasi terbaik ditemukan pada perlakuan C (50%) yang menghasilkan rata-rata kadar asam sitrat sebesar 1,66 ± 0,78 mg/L dengan kadar puncak mencapai 3,08 mg/L pada waktu fermentasi jam ke-108. Penurunan pH hingga mencapai 3,0 pada variasi C mengonfirmasi efisiensi sekresi asam organik yang lebih tinggi. Kesimpulan penelitian menunjukkan bahwa konsentrasi limbah mangga 50% merupakan dosis optimal untuk biokonversi asam sitrat.
